Serum IgA2 is the second subclass of Immunoglobulin A in humans and is more abundant at mucosal surfaces than in circulation. Structurally, IgA2 lacks the extended hinge region present in IgA1, making it more resistant to cleavage by bacterial proteases and particularly well suited for mucosal immune defense.
Functionally, IgA2 contributes to immune protection by neutralizing pathogens, facilitating immune complex clearance, and engaging FcαRI receptors on neutrophils and monocytes to support antibody-dependent cellular responses. Its structural resilience allows IgA2 to remain active in environments rich in microbial proteases, such as the gastrointestinal and respiratory tracts.
IgA2 plays an important role in mucosal immunity and host–microbiome interactions, particularly within the intestinal tract. Alterations in IgA2 levels or function have been associated with inflammatory bowel disease, chronic mucosal infections, and dysregulated mucosal immune responses.
Clinically, IgA subclass measurements—including IgA2—can support evaluation of selective IgA deficiencies, mucosal immune dysfunction, and immunodeficiency disorders. In secretory IgA, IgA2 represents a substantial fraction of total IgA, often approaching parity with IgA1, reflecting its specialized role in protecting mucosal surfaces from microbial challenge.
IgA2 is widely used in research and in vitro diagnostics to study mucosal immunity, host–microbiome interactions, and antibody effector mechanisms. Due to its resistance to bacterial proteases, IgA2 is particularly valuable for investigating immune responses in the gastrointestinal and respiratory tracts. In the IVD industry, purified IgA2 is commonly used as an immunogen for antibody production and as a reference material for calibrators, controls, and subclass-specific immunoassays that measure IgA distribution and immune function.
